高糖对人肾小球系膜细胞MPO、GSH-ST活性和ICAM-1表达影响及茶多酚的干预作用 被引量:1

摘要 目的探讨高糖对人肾小球系膜细胞髓过氧化物酶(MPO)、谷胱甘肽-硫转移酶(GSH-ST)活性和细胞间黏附分子1(ICAM-1)表达的影响及茶多酚对其的干预作用。方法应用分光光度比色法检测高糖作用后系膜细胞上清液中MPO、GSH-ST活性及茶多酚对其活性的改变;免疫细胞化学法和Western印迹检测高糖作用后系膜细胞内ICAM-1蛋白的表达及茶多酚对其表达的影响。结果高糖使系膜细胞上清液中MPO活性增加,GSH-ST活性降低,使系膜细胞内ICAM-1表达增加;茶多酚可显著改善高糖对系膜细胞的影响。结论高糖导致人肾小球系膜细胞氧化失衡,增强ICAM-1的表达;茶多酚可有效干预高糖的作用。 Objective To investigate the effect of high glucose on the production of GSH-ST, MPO and ICAM-1 in human mesangial cells and the intervention of tea polyphenols (TPs). Methods The mesangial cells were cultured in the high glucose with or without TPs up to 36 hours. The activities of GSH-ST and MPO in the supernatants were detected by spectrophotometry. The protein expression of ICAM-1 was measured by immunocytochemistry and Western blot. Results MPO increased, GSH-ST decreased, and ICAM-1 increased in the high glucose media without TPs. However,MPO decreased, GSH-ST increased, and ICAM-1 reduced in the high glucose media with TPs. Conclusion High glucose could induce the mesangial cells to produce more reactive oxygen species and ICAM-1. TPs could inhibit the effect of high glucose on the mesangial cells.
