盐酸氨溴索、地塞米松对大鼠胎肺TGF-β_3表达的影响 被引量:1

摘要 目的比较产前给药盐酸氨溴索、地塞米松对大鼠胎肺转化生长因子-β3(TGF-β3)基因和蛋白表达的影响。方法9只孕鼠随机分为生理盐水对照组、地塞米松组、盐酸氨溴索组,每组3只。孕16、17、18d腹腔注射给药3剂。孕19d取胎鼠肺,RT-PCR法检测TGF-β3基因表达水平;免疫组织化学法检测TGF-β3蛋白表达水平。结果地塞米松组和盐酸氨溴索组TGF-β3基因和蛋白的表达均明显高于对照组(P<0.05)。地塞米松组TGF-β3基因和蛋白的表达高于盐酸氨溴索组(P<0.05)。结论产前给药盐酸氨溴索、地塞米松均能促进胎肺发育相关因子TGF-β3基因及蛋白的表达,从而促进胎肺功能的发育。 Objective To compare the effects of antenatal ambroxol and dexamethason on expression of transform growth factor-β3 (TGF-β3) in rat fetal lung. Methods Nine pregnant rats were divided into three groups randomly with 3 rats each: ambroxol group, dexamethasone group and control group. Three doses were given on gestational day 16,17 and 18 by intraperitoneal injection. On gestational day 19, rat fetal lungs were collected. The expression level of TGF-β3 mRNA was measured by RT-PCR. The expression level of TGF-β3 protein was measured by immunohistochemistry. Results The expression levels of mRNA and protein of TGF-β3 in the therapy groups were higher than those in control group(P〈0. 05). The expression level of mRNA and protein of TGF-β3 in dexamethasone group was higher than that in ambroxol group(P〈0. 05). Conclusion Ambroxol and dexamethasone could promote the expression of mRNA and protein of TGF-β3 in rat fetal lung.
