桂枝汤对脾虚大鼠NFAT mRNA,IL-4 mRNA,IFN-γ mRNA的干预作用 被引量:7

摘要 目的:采用RT-PCR方法来探讨桂枝汤对脾虚NFAT mRNA,IL-4 mRNA,IFN-γ mRNA的影响以及干预Th1/Th2细胞漂移的机制.方法:40只大鼠随机分为空白对照组(A组)、脾虚组(B组)、桂枝汤大剂量组(C组)和桂枝汤小剂量组(D组)等4组,每组10只;用规范的脾气虚证大鼠模型造模方法将B,C和D组大鼠造模,B,C和D组分别用蒸馏水、桂枝汤大剂量、桂枝汤小剂量灌胃,标本采集后用RT-PCR检测NFAT mRNA,IL-4 mRNA和IFN-γmRNA表达水平.结果:脾虚组的NFAT mRNA,IL-4 mRNA表达上调;而IFN-γ mRNA表达下调,经桂枝汤大、小剂量组治疗后,NFAT mRNA,IL-4 mRNA表达下调,IFN-γ mRNA表达上调.结论:桂枝汤对Th1/Th2细胞漂移作用途径可能是通过下调NFAT mRNA的表达,恢复了IFN-γ mRNA正常转录水平,进而下调IL-4 mRNA的表达,使脾虚大鼠Th1/Th2细胞达到一种新的平衡. AIM: To explore the intervention of Guizhi decoction on NFAT mRNA, IL-4 mRNA, IFN-γ mRNA and the mechanism for Th1/Th2 cell shifting. METHODS: Forty rats were randomly divided into 4 groups : normal group ( group A), asthenic spleen group(group B), high-dosage group (group C) and low-dosage group (group D) with 10 rats in each group. The asthenic spleen rat models were established in group B, C and D. Rats in group B, C and D were fed with normal distilled water, high-dosage of Guizhi decoction and low-dosage of Guizhi decoction, respectively. NFAT mRNA, IL-4 mRNA, IFN-γ mRNA were determined by RT-PCR. RESULTS: Expressions of NFAT mRNA, IL-4 mRNA in group B were raised, however, the expression of IFN-γ mRNA was decreased; after treatment, the expressions of NFAT mRNA, IL-4 mRNA in group C and D were decreased and the expression of IFN-γ mRNA was raised. CONCLUSION: Guizhi decoction may interfere Thl/Th2 cell shifting through down-regulating NFAT mRNA, recovering normal transcription of IFN-γ mRNA, down-regulating expression of IL-4 mRNA, and reaching a new balance of Th1/Th2 cell.
