摘要
目的探讨手掌参醇提物(GcEE)对染矽尘大鼠肺组织Ⅰ、Ⅲ型胶原合成的影响,阐明其抗矽肺纤维化的作用。方法32只大鼠随机分成对照组、染矽尘组、GcEE治疗组和汉防己甲素(TT)组。建立矽肺动物模型后第2天起,GcEE治疗组、TT治疗组分别灌胃给予GcEE[8g/(kg.d)]、TT[50mg/(kg.3d)]治疗,对照组和染矽尘组给予等容蒸馏水,给药28d后处死大鼠,观察肺组织Ⅰ、Ⅲ型胶原合成,并用图像分析系统进行定量分析。结果GcEE可显著降低染矽尘大鼠肺系数,减少肺组织Ⅰ、Ⅲ型胶原合成。结论GcEE能够明显抑制染矽尘大鼠的肺纤维化。
Objective To study the effects of Gymnadenia conopseaethanol extract (GcEE) on the synthesis of types Ⅰ and Ⅲ codagen in lungs of rats exposed to silica. Methods Thirty - two rats were randomly divided into four groups: control group, silica group, GcEE - treated group, and tetrandrine - treated group. Silicotic rat model was developed by direct tracheal instillation of silica into rat lungs. From the second day of model development,rats in GcEE group and in tetrandrine group were orally given GcEE[8 g/(kg·d) ] and tetrandrine[ (50 mg/ (kg·3d)], respectively, meanwhile the other two groups were orally given distilled water. After twenty- eight days, rats in each groups were sacrificed. Paraffin - embedded lung sections were stained with Sirius red,and detected for collagens Ⅰ, Ⅲ by polarization microscopy and Image - Pro Plus version 4.5 for WindowsTM. Results GcEE significantly reduced the lung/body ratio of rats exposed to silica and decreased the Ⅰ and Ⅲ collagen synthesis induced by silica in lung. Conclusion GcEE has the effects on ameliorating the silica- induced pulmonary fibrosis.
