摘要
目的:讨论N-钙黏素融合蛋白N-cad-Fc对P19细胞分化过程中伴随的细胞凋亡的影响。方法:分子生物学技术制作N-钙黏素融合蛋白N-cad-Fc。视黄酸(retinoic acid,RA)诱导P19细胞分化,培养于明胶(gelatin)包被培养板上的P19细胞为对照组,普通光学显微镜观察其神经分化情况,TURNEL法和DNA片段化分析法检测细胞凋亡情况,alamarBlueTM染色法检测细胞存活情况。结果:培养于N-cad-Fc包被培养板上的P19细胞经RA诱导分化后细胞形态与明胶包被培养板上P19细胞分化形态一致,但分化过程中伴随的凋亡现象被明显抑制,分化诱导后细胞的存活率大大提高。结论:使用N-cad-Fc融合蛋白培养技术可以在不影响细胞分化状态的前提下降低分化过程中细胞的死亡率,为开发低死亡率的高效细胞分化诱导方法创造了条件。
Objective: To study the effects of N-cad-Fc on apoptosis accompanied by P19 differentiation process. Methods: N-cad-Fc was made through biomolecular technology. P19 differentiation was induced by retinoic acid (RA), and P19 cells cultured on gelatin were used as control. Differentiated cells were observed with microscope. TURNEL method and DNA fragmentation analysis were used for apoptosis confirmation. Survival .cells were detected by AlamarBlueTM method. Results: P19 cells cultured on N-cad-Fc coated dishes showed the same differentiation morphology to those cultured on gelatin coated dishes, but apoptosis accompanied by differentiation process was obviously inhibited and cell viability was largely improved. Conclusion: With N-cad-Fc culture method, cell death ratio could be abased without affecting differentiation process. This method might provide us a low death ratio and high efficiency way for inducing cell differentiation.
