羟基积雪草苷对LPS致小鼠痛觉增敏影响研究 被引量:4

摘要 目的:观察羟基积雪草苷对小鼠腹腔注射脂多糖(LPS)诱导痛觉增敏型疼痛的影响,并探讨羟基积雪草苷对髓过氧化物酶(MPO)、白介素-6(IL-6)和α-肿瘤坏死因子(TNF-α)的影响。方法:随机将小鼠分为空白组、模型组、盐酸度冷丁组、羟基积雪草苷10、20、40mg·kg-1六组,通过腹腔注射LPS100μg·kg-1建立痛阈增敏型疼痛模型,采用醋酸扭体法、热板法观察羟基积雪草苷镇痛效应;通过MPO试剂盒检测小鼠脑组织MPO活性、ELISA法检测血浆TNF-α,IL-6含量的变化。结果:羟基积雪草苷10、20、40mg·kg-1各组明显减少醋酸扭体次数,提高小鼠痛阈。降低脑组织MPO活性和血浆TNF-α、IL-6含量。结论:MC具有明显镇痛效应,其镇痛效应可能与降低MPO、IL-6和TNF-α有关。 Objective: To observe the analgesic effect of medecassoside(MC) on the model of hyperalgesia induced by intraperitoneal LPS in mice,and investigate the change of MPO,IL-6 and TNF-α.Methods'Mice were randomly divided into normal group,model group,dolantin group and MC 10,20,40mg·kg^-1 group.Hyperalgesia was induced by intraperitoneal LPS 100μg·kg^-1 in mice.The analgesic effect of MC was assessed by the hot-plate stimulation and the writhing test induced by acetic acid ;the activity of MPO in brain tissue was tested by MPO kits; the content of TNF-α,IL-6 in plasma was tested by ELISA. Results: MC10,20,40mg·kg^-1 could signifi- candy reduce the number of writhing, increase pain threshold,decrease the activity of MPO and the content of TNF-α ,IL-6. Conclusion: MC has the effect of analgesia and its effect may be related with inhibiting MPO, IL- 6 and TNF-α.
