新Y染色体DYS726在山西汉族人群中遗传多态性 被引量:2

摘要 目的对新的DYS726基因座的基因多态性和法医学意义进行研究。方法山西汉族人群盅随机筛选110名男性,提取静脉血DNA,用基因数据库(GDB)中的引物进行聚合酶链反应(PCR)扩增,银染显色。对其筛选的等位基因测序,构建等位基因分型标准物;按照国际法医学会(ISFG)原则命名各等位基因。结果首次选择了Y染色体基因座DYS726,该基因座为4核苷酸简单重复STR。重复单位为CTTC。在110名山西汉族男性个体血样中共检出了4个等位基因。基因频率分别为11-0.0273;12-0.5455;13-0.1818;14-0.2455。基因多样性为0.6139,在法医学应用方面,其个人识别能力(DP)和非父排出率(PE)均为0.6139。结论新Y染色体基因座DYS726具有较高的遗传多态性,在法医学及人类遗传学方面具有较高应用价值。 Objective To investigate the gene polymorphism and the utility of a new locus DYS726. Methods Each allele of this Y-STR loci was sequenced and the allele ladders were constructed. Alleles of this Y-STR locus were nominated according to reconmaendations of the International Society of Forensic Genetics. Results We first choose the Y-STR locus DYS726. The number of the simple repeat nucleotide is four and the sequence is CTTC. Four alleles had been found in 110 unrelated male venous blood in Chinese Han population of Shanxi. The frequencies of throe locus are 11 - 0. 027 3 ; 12 - 0. 545 5 ; 13 - 0. 181 8 ; 14 - 0. 245 5. If used in forensic science, both of the power of discrimination (PD) and the power of exclusion (PE) were 0. 6139. Conclusion DYS726 has a high level of genetic polymorphism and can be used in forensic science and human genetics research.
