凝血酶预处理减轻凝血酶所致的脑水肿 被引量:1

摘要 目的:探讨凝血酶预处理对脑水肿的影响,检测肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)在脑组织内的表达并分析其与凝血酶所致脑水肿的关系。方法:建立大脑皮质内血肿模型。选择健康成年SD大鼠40只,随机分为作为对照的未预处理组(ST组)和小剂量凝血酶(1U)预处理组(TT组)。观察不同时相脑组织含水量及钠含量的变化及TNF-α在脑组织内的表达。结果:ST组脑组织含水量和钠含量明显高于TT组,且第1天高于第3天。TT组与ST组脑组织内TNF-α阳性表达与含水量的变化相一致。结论:凝血酶预处理可以减轻凝血酶所致的脑水肿;TNF-α的表达与凝血酶所致的脑水肿有关。 Objective To explore the effect of the cerebral thrombin preconditioning on the thrombin-induced brain edema,to detect the expression of tumor necrosis factor-α(TNF-α),and to analyse the relationship between TNF-α and the thrombin-induced brain edema.Methods Forty SD rats were randomly divided into a ST group and a TT group.The rats received 50 L saline(ST group)or 1U thrombin infusion(TT group),and received the second infusion(10U thrombin)24 h later.The rats were sacrificed at 24 and 72 h after the second infusion in order to examine the changes of brain water and sodium contents as well as the expression of TNF-α in the brain.Results The brain water and sodium contents in the ST group were significantly higher than those on the TT group,and those on the 1st day were higher than those on the 3th day.The positive expression of TNF-α and in the change of water content were identical in the TT group and the ST group.Conclusion Thrombin preconditioning can alleviate the thrombin-induced brain edema.The increase of TNF-α expression after thrombin treatment may be related to the thrombin-induced brain edema.
