摘要 研究了用高效液相色谱法快速测定桑叶中的糖类物质的方法。色谱泵为LC-10ATvp高压恒流泵。检测器为RID-10A示差折光检测器,色谱柱为Agilent ZORBAX NH24.6×250mm,柱温40qc,检测池温度为40℃,流动相为乙腈:水=75-25,流速1.0mL/min,进样体积10曲。结果表明,此方法测定各种单糖的稳定性好,用相对标准偏差RSD表示,鼠李糖3.33%,木糖0.28%,果糖0.62%,葡萄糖0.34%,蔗糖3.41%,麦芽糖2.26%,乳糖3.09%,棉子糖0.19%。该方法快速简捷,精密度高,重现性好。 Determination of carbohydrates in mulberry leaves was studied by HPLC. The system of chromatography included the high-press constant-current of LC-10ATvp,the refractive index detector, RID-10A ,and the chromatographic column of Agilent ZORBAX NH2 4.6×250mm. The terperature in column and tetector was 40℃. The flowing phase was CHa CN:H2O=75:25,with a flow rate of 1.0 mL/min. The input volume was 10μL. The result showed that the stability of rhamnose, xylose, fructose, glucose, sucrose, maltose, lactose and raffinose were 3.33%, 0.28%, 0. 62%,0.34% ,3.41% ,2.26% ,3.09% and 0.19% respectively. The method was accurate,rapid,simple and reproducible.
