动物性食品中奇异变形杆菌PCR检测方法的研究 被引量:11

摘要 研究制定动物源性食品中奇异变形杆菌的PCR检测方法,为日常食品卫生监测提供快速检验的依据。根据奇异变形杆菌抗亚碲酸盐基因的保守序列设计Q248F/R,确定PCR条件,利用标准菌株检验方法的特异性和敏感度。引物Q248对奇异变形杆菌检测具有良好的特异性,对人工布菌样品的检测敏感度达1.2×102cfu/100g。通过对扩增目的片段的核酸序列测定,目的片段大小为248bp。该方法用于奇异变形杆菌检测具有特异性强、敏感、快速的特点,适用于动物性食品中奇异变形杆菌检测的快速筛选检验。 To establish Polymerase Chain Reaction (PCR) method for detection Proteus mirabilis, so as to provide a reference method for sanitary inspection. On the basis of the conserved section of the tellurite resistance gene sequence of Proteus mirabilis, a sets of specific primers was designed, a PCR method developed to identify. Proteus mirabilis. The PCR method established in this article are highly specific, and it can detect Proteus mirabilis 1.2 × 10^2 cfu/100g dried meat in sliced foml by amplifying a 248 bp DNA fragment.The PCR assay was rapid,reliable, specific and sensitive,and the method can be used in the detection of Proteus mirabilis in animal- derived food.
