能谱-X线微束分析检测α-actinin在额叶皮质神经干细胞定向分化神经元的表达 被引量:1

摘要 目的用能谱-X线微束分析检测新生大鼠额叶皮质神经干细胞分化的神经元各时段α辅肌动蛋白(α-actinin)原位超微分布及量的表达变化。方法采用神经干细胞培养、Aclar膜电镜免疫细胞化学方法、能谱-X线微束分析术对各时段分化的神经元中α-actinin的表达进行定位、定量检测。结果电镜免疫细胞化学显示α-actinin阳性表达呈电子密度高的球形颗粒,神经干细胞向神经元分化过程中,其表达分布逐渐密集由核周至充满胞质并伸至突起中,随着突起伸展而延伸分布,在原位超微结构表达基础上,能谱-X线微束分析检测结果表明了随着神经元的分化成熟,α-actinin的表达量逐渐增加。结论大鼠额叶皮质神经干细胞向神经元分化各时段α-actinin表达的分布变化及量的增加均与分化的神经元的生长发育成正相关。 Objective To determine the position ultramicro-distribution and the quantity expression changes of α-actinin in the neurons dif- ferentiated from temporal lobe cerebral cortex nerve stem cells in newborn mouse at various time intervals with energy dispersive analysis system of X-my. Methods Neural stem cells were cultivated, and the localization and the quantity of α-actinin expression in differentiated neurons at different time points were determined with Aclar membrane EM-immuno-cytochemistry and energy dispersive analysis system of X- ray. Results EM-immano-cytochemistry showed that α-actinin positive expression assumed the high electronic density sphere pellet. During the differentiation from the nerve stem cells to the neurons,its expression distribution gradually became denser,extending from the perinuclear,full into the plasma and to the dendrite,along with dendrite extension. It was found that the quantity increased gradually during the differentiation and maturation of neurons on the base of position ultra-structure expression with energy dispersive analysis system of X-ray. Conclusion The distribution change and the quantity increase of α-actinin expression in the neurons differentiated from mouse temporal lobe cerebral cortex nerve stem cells at various time intervals show a positive correlation with the neuron growth.
