摘要 目的探讨温度对PGC-1表达的影响,旨在明确PGC-1的表达对心肌的影响,找出一种新的心肌保护方法。方法选用30只Wistar大鼠雌雄随机分为3组,即正常组(1组)、对照组(2组)、低温组,冰冻切片后行免疫组化染色并检测染色密度。电镜的超微结构检查后观察线粒体改变。结果经过分析得出正常组与对照组无显著性差异,而正常组、对照组与实验组均有显著性差异。正常组及对照组心肌组织的超微结构没有发生明显的改变,但低温组中随温度的降低线粒体的数目有所增加。结论低温可以使PGC-1的表达增强,从而使线粒体增殖并加强线粒体的氧化供能作用;在低温下,心肌的线粒体会有所增殖,为心肌的运动提供更多的能量,加速心肌的代谢。 Objective To definite the effect of temperature to PGC-1, to definitude the relation between the myocardium cells and the PGC- 1, so that we can find a new way to protect the myocardinm. Methods 30 Wistar mice were randomly divided into 3 groups: ① heart in vivo ② heart in vitro ③ low temperature groups,cut of the two pieces of myocardium of left ventricle,one operating under -70℃ in order to observe immunohistochemistry (IH).Afterward,the second of the myocardium was put into 2.5% glutaral solution immediately ready for the examination of ultrastructure. Results There were no significant differences between common group and controll group .But,there were significant differences shown at hypothermic group compared with controlling group.there were no significant differences in ultrastructure between controlling group and common group.but,In the low temperature irrigated groups,the more low temperature is,the more chondriosomes have shown. Conclusion Low temperature can stregthen the chondriosomes oxide activities,the expression of PGC-1 changes to be obviously.In the low temperature, the myocardium chondriosomes will proliferate, supply more ATP for metabolism.
