Resolution of DL-Arginine by Cell Immobilization of Arginine Deiminase

摘要 以DL-精氨酸为原料,用固定化细胞酶法拆分DL-精氨酸,并对拆分条件进行了研究。结果表明:最适反应温度55℃,最适pH=6.0,c(DL-精氨酸)=0.3 mol/L,ρ(菌体)=30 g/L,拆分反应10 h,拆分率达98%以上。连续反应10批次,固定化细胞仍保留最高酶活力的75%。产物经分离纯化,D-精氨酸收率达84.0%,[α]2D5=-27.4°〔ρ(D-精氨酸)=20 g/L,5 mol/L盐酸〕。 An enzymatic method for optical resolution of DL-arginine by immobilized cell was developed, and many factors affecting the resolution were studied. The result shows that the optimum conditions are : 55℃ , pH = 6.0, c (DL-arginine) = 0. 3 mol/L and p (cell) = 30 g/L. The conversion rate of L-arginine is higher than 98%. After ten times repeated batch operation,the relative activity of immobilized cell was still 75%. The products were separated and purified on ion-exchange resin. The yield of D-arginine was 84.0%.
