加味黄芪建中汤对脾气虚证大鼠胃泌素基因表达的影响 被引量:10

摘要 目的:探讨加味黄芪建中汤不同浓度、不同时间对脾气虚证大鼠胃泌素(gastrin,GAS)基因表达的影响。方法:以大黄法、游泳力竭法、饥饿法三因素复合的方法复制脾气虚证大鼠模型,造模大鼠随机分成模型组、加味黄芪建中汤低、中、高剂量组,在用药后不同时间(7d、14d、21d),用RT-PCR方法检测加味黄芪建中汤对胃窦GAS mRNA表达影响。结果:与模型组比较,加味黄芪建中汤中、高剂量组血清GAS水平升高(P<0.05,P<0.01)、胃窦GAS mRNA表达上调(P<0.05,P<0.01);中、高剂量组比较,差异有统计学意义(P<0.05),以中剂量组为佳。结论:加味黄芪建中汤能上调GAS mRNA表达可能是其治疗脾气虚证的机制之一。 Objective: To explore the effect of modified HuangqiJianzhong decoction (MHQJZD) with various concentrations at different time points on gastrin(GAS) gent expression level in spleen- qi deficiency rats. Methods: Spleen-qi deficiency syndrome was induced in rats by compound methods which including Radix ct Rhizoma Rhci, exhausting and hunger. Splecn-qi deficiency rats were randomly divided into the model control group and the low, medium and high dosage of MHQJZD group. Reverse transcriptase polymerase chain reaction was used to observe the effect of MHQJZD on the GAS mRNA of gastric antrum in rats treated at different time points (7, 14, 21d). Results: As compared with that in model control group, the GAS gent expression level were both significantly higher in the medium and high dosage groups( P 〈 0.05, P 〈 0.01 ). And the increment in the medium dosage group was significantly higher than that in the high dosage group( P 〈 0.05). Conclusion: The regulation on GAS gone expression is possibly one of the mechanisms of MHQJZD for treating spleen- qi deficiency.
