摘要
目的探讨睫状神经营养因子(CNTF)蛋白在慢性高眼压性青光眼大鼠视网膜中的定位及表达情况。方法采用水下电凝巩膜静脉建立大鼠慢性高眼压性青光眼模型,在1、3、7、14、28d分别摘取眼球,运用免疫组织化学和Westernblot法检测大鼠视网膜CNTF蛋白的定位及表达变化。结果对照组大鼠视网膜神经节细胞(RGCs)层有少量CNTF蛋白表达,青光眼大鼠视网膜CNTF蛋白的表达显著增加,建立模型后第7~14d表达量达到高峰,此时除了神经节细胞层外,内外核层亦发现有CNTF蛋白的表达,之后表达减少。结论青光眼大鼠视网膜内源性CNTF表达增加,可能与促进损伤的RGCs的存活和轴突再生密切相关。
Objective Nowadays ciliary neurotrophic factor(CNTF) is believed to promote the growth and development and stimulate the regeneration of axons of retinal ganglion cells(RGCs) and therefore consequently obtain a great deal of attention in the field of ophthalmology. The objective of this study was to detect the expression of CNTF protein in the retina of glaucoma model with increased intraocular pressure(IOP). Methods The glaucoma model was created by electrocoagulating at least 3 episcleral and limbal veins in 36 adult Sprague-Dawley rats. The rats were divided into pseudosurgery group (6 rats) and chronic hypertention group(30 rats), lOP was measured in 1,3,7,14 and 28 days after modeling. The location and expression of CNTF protein were detected at 1,3,7,14 and 28 days after surgery by immunohistochemistry assay and Western blot analysis. Results The lOP was moderately elevated after modeling, showing a significant difference compared with pseudosurgery group in various time points(P 〈 0. 01 ). A minimum protein expression of CNTF was found only in the ganglion cell layer of the retinas of the pseudosurgey group. In model group,CNTF was expressed in retinal inner nuclear layer and outer nuclear layer. The expression level of CNTF reached peak in the 7th day after surgery and reduced 28 days later. Conclusion The increased protein expression of endogenous CNTF in the retina of a rat glaucoma model is closely linked to the axonal reaction of RGCs after the induction of ocular hypertension.
