角质细胞生长因子对无排卵型功血子宫内膜上皮细胞生长与分泌CA125的影响 被引量:3

摘要 目的观察角质细胞生长因子(keratinocyte growth factor,KGF)对离体培养的无排卵型功血子宫内膜上皮细胞生长和分泌CA125的影响。方法体外培养无排卵型功血子宫内膜上皮细胞,加入不同浓度的KGF作用后,微量噻唑蓝分析法检测细胞量,了解生长情况,磁酶免法测定CA125分泌,分析两者之间的相关性,并与培养的正常子宫内膜上皮细胞进行比较。结果无排卵型功血子宫内膜上皮细胞与正常子宫内膜上皮细胞的外观特征相似;CK19免疫组化染色结果为无排卵型功血子宫内膜上皮细胞的细胞质被染成棕黄色。②不同浓度的KGF作用体外培养的无排卵型功血子宫内膜上皮细胞后,细胞增生明显,但增生程度不如正常子宫内膜上皮细胞(P<0.05)。③不同浓度KGF作用后两组子宫内膜上皮细胞的CA125浓度均升高,无排卵型功血子宫内膜上皮细胞的增高程度明显低于正常子宫内膜上皮细胞(P<0.05)。④正常和功血的子宫内膜上皮细胞中二者的吸光度值与细胞培养上清液中的CA125浓度有显著的相关性,两者的相关系数分别为0.681和0.885(P<0.05)。结论KGF对体外培养的无排卵型功血子宫内膜上皮细胞有促进生长和分泌CA125的作用,但促进作用的程度不如正常子宫内膜上皮细胞,提示无排卵型功血子宫内膜上皮细胞存在与正常子宫内膜上皮细胞不同的生长机制和功能,可能参与无排卵型功血的发病。 Objective To observe the effect of keratinocyte growth factor (KGF) on growth and CA125 secretion of cultured endometrial epithelial cells (EECs) from the patients of anovulatory dysfunctional uterine bleeding (ADUB). Methods The EECs from 30 cases of ADUB and 22 cases of resected uterus due to other diseases but with normal menstruation were separated and cultured with KGF of different concentration. Their growth curves were drawn by M3T method. Quantity of CA125 in EECs was determined. Results EECs from ADUB patients appeared similar to normal EECs and were CK19 positive. KGF increased the proliferation of EECs from ADUB patients significantly ( P 〈 0.05 ), but the proliferation degree was lower than that of normal EECs. KGF increased the CA125 secretion by EECs from ADUB patients significantly (P 〈0.05), but the degree of CA125 secretion was lower than that of normal EECs. The proliferation of EECs was correlated significantly with CA125 secretion (P 〈 0.05 ). Conclusion KGF enhances the growth and CA125 secretion of cultured EECs from ADUB patients, but the degree in EECs from ADUB patients was lower than that of normal EECs. These suggest that EECs from ADUB patients have different growth mechanism with normal EECs.
