含黑色素细胞的表皮替代物构建及对色素脱失的疗效 被引量:3

摘要 目的联合应用黑色素细胞和表皮细胞构建组织工程化表皮替代物,并应用于色素脱失治疗。方法利用幼儿包皮分离、培养和扩增出表皮细胞和黑色素细胞,接种于壳聚糖明胶膜,构建成含黑色素细胞的表皮膜片。建立裸鼠动物模型,将含黑色素细胞的表皮膜片移植于治疗组的受区,同时设立表皮细胞膜片为对照组。术后定期行大体观察、组织切片HE染色、抗S-100蛋白免疫组化染色,以检测黑色素细胞在创面的成活情况。结果联合应用人黑色素细胞和表皮细胞接种于壳聚糖/明胶膜能够构建出含黑色素细胞的表皮替代物,并应用于动物实验,检测出黑色素细胞成活良好。结论联合应用人黑色素细胞和表皮细胞接种于壳聚糖/明胶膜的表皮替代物能使色素脱失的动物模型恢复色素功能,对色素脱失具有良好的治疗效果。 Objective To construct the pigmented epidermal substitute by human epidermal cells and melanocytes for treating hypopigmentation. Methods Epidermal keratinocytes and melanocytes were harvested from human foreskin, and were cultured and purified and then seeded onto chitosan/gelatin membrane. The pigmented epidermal substitute was used in vivo. Athymic mouse as animal model, the group of treatment was covered with the pigmented epidermal substitute, and the epidermal substitute without melanocytes was grafted on the control group. The pigmented epidermal substitute and its effect on hypopigmentation were evaluated with gross observation, histological examination with HE staining and anti S-100 protein immunohistochemistry. Results The pigmented epidermal substitute could be constructed with cultured human epidermal cells, melanocytes and chitosan/gelatin membrane. The epidermal substitute was applied in animal experiment, and human melanocytes were detected in receptor site. Conclusion The melanocytes and chitosan/gelatin membrane can be constructed and have good clinical application in pigmentation recovery.
