摘要
目的:通过结扎冠状动脉造成大鼠急性心肌缺血损伤模型,探讨复方黄芪保心汤对缺血心肌心功能及心肌细胞超微结构的影响。方法:结扎冠状动脉造成大鼠急性心肌缺血损伤模型,以目前治疗冠心病疗效确切的复方丹参滴丸作对照,试图观察黄芪保心汤对缺血心肌心功能(病理生理)及心肌超微结构(病理解剖)的影响。结果:实验性心肌缺血大鼠的心功能明显下降,心肌结构改变显著,黄芪保心汤和复方丹参滴丸均能增强大鼠的心肌收缩功能,改善心肌形态学和细胞超微结构的改变,与模型组相比,有显著性差异(P<0.05,P<0.01),但二者相比,差异无统计学意义(P>0.05)。结论:以益气活血化痰立法的黄芪保心汤具有良好的抗心肌缺血作用,其作用机理可能与增强心肌收缩力、抑制心肌细胞的坏死和凋亡有关,提示中药复方制剂在防治心肌缺血方面大有潜力。
Objective: To observe the effect of Astragalus Heart- Protecting Decoction(AHPD) in prevention and treatment of acute myocardial ischemia(AMI) induced by ligation of coronary artery (LCA). Method: 48 rats were randomly divided equally into four groups, group A was treated with Compound Danshen Dripping Pills( CDDP), group B was treated with AHPD, group C and group D were seperately regarded as control group and sham - operated group. 2 weeks later, models of rats with AMI were made by LCA. After 3 hours, Left ventricular systolic pressure ( LVSP), dp/ dtmax and the changes of myocardial histopathology were observed. Results: After treatment, the levels of LVSP,dp/dtmax were significantly higher than that in the control group( P 〈 0.05 or P 〈 0.01 ), furthermore, the changes of myocardial histopathology were statistically better than that in the control group , As compared with CDDP group, there was no statistical difference between them ( P 〉 0. 05 ). Conclusion: AHPD can act against myocardial ischemia effectively by means of enforcing Qi,activating blood and deleting spider, one of the preventive and therapeutic mechanisms of which may be related with enhancing myocardial systolic power and resisting(sustaining) the necrosis and oppotosis of myocardial cells ,which shows the splendid future of Traditional Chinese Medicine in preventing and treatment of acute myocardial ischemia.
