电针对慢性应激大鼠开场行为与认知功能的影响 被引量:1

The effects of electroacupuncture on open-field behavior and recognitive function of the chronic stress model rats

摘要 目的观察电针与氟西汀对大鼠抑郁行为和认知功能的影响,探讨其作用的不同机制。方法采用孤养结合慢性温和应激大鼠模型,对不同组大鼠进行电针与氟西汀干预。以体质量为指标进行大鼠一般状况评价;运用开场实验评定大鼠行为改变;运用Morris水迷宫成绩评定大鼠认知功能改变。结果21d后,正常组、模型组、电针组和氟西汀组水平运动次数分别为(24.1±7.05)次、(10.1±4.6)次、(17.3±8.21)次、(11.8±4.13)次,其中模型组、氟西汀组与正常组比较差异有极显著性(P〈0.01),电针组与正常组比较差异有显著性(P〈0.05);各组间垂直运动差异无显著性(P〉0.05);正常组、模型组、电针组和氟西汀组Morris水迷宫潜伏期分别为(10.46±2.57)s、(30.27±3.56)s、(21.16±4.86)s、(17.76±5.78)s,其中模型组、电针组、氟西汀组与正常组相比均显著增加(P〈0.01),电针组、氟西汀组与模型组相比持续时间下降差异有极显著性(P〈0.01)。结论电针与氟西汀皆可减轻慢性应激模型大鼠体质量下降,改善其行为与认知功能抑制,但二者作用各有优势,提示其机制可能不同。 Objective To investigate the effect of the electroacupuncture and fluoxetine on the behavior and recognitive function in chronic stress model rats. Methods 48 rats were divided into four groups, the control group, model group, EA group, and fluoxetine group. Then the rats were subjected to successive 21 days of seven different ways of chronic stress. Take electroacupuncture and fluoxetine as treatment method. Using weight,openfield test and Morris water maze test to evaluate the curative effect. Results Eleetroaeupuneture and fluoxetine can significantly alleviate the weight loss of the model rats. The rearing times in open-field test of the four groups orderly are 24.1 ±7.05, 10.1±4.6, 17.3 ± 8.21, 11.8 ±4.13. The model group and the fluoxetine group have a very significant difference compared to the control group( P〈 0.01 ) ,while the EA group has a significant difference compared to the control group. There are no significant difference between the four groups in the crossing time in the open-field test. The latent period in Morris water maze test of the four groups orderly are 10.46 ±2.57, 30.27 ±3.56,21.16 ±4.86,17.76 ±5.78. Compared to the control group, the latent period of the model group, EA group and the fluoxetine group increased significantly( P 〈 0.01 ), and compared to the model group, the EA group and the fluoxetine group decreased significantly ( P 〈 0.01 ). Conclusion Chronic stress can induce the decrease of the activities and exploring behaviors of the model rats. Eleetroaeupuneture and fluoxetine can increase the activities and exploring behaviors of the model rats. The effect of the eleetroaeupueture and fluoxetine may have different emphasis and their mechanism may be not the same.
