佐剂关节炎大鼠血小板参数和细胞因子的变化及其相关性研究

The changes of platelet parameters,cytokines and the research of correlation in adjuvant arthritis rats

目的观察佐剂关节炎(adjuvant arthritis,AA)大鼠血小板参数(血小板计数PLT、血小板压积PCT、血小板平均体积MPV、血小板分布宽度PDW)、细胞因子(IL-1β、TNF-α、IL-10)的变化并对其行相关性分析。方法将16只SD雄性大鼠随机分为健康对照组、模型组,每组8只,用弗氏完全佐剂(Freund's com-plete adjuvant,CFA)向模型组大鼠右后足跖皮内注射0.1ml诱发大鼠产生关节炎,观察两组大鼠血小板参数(PLT、PCT、PDW、MPV)和细胞因子(IL-1β、TNF-α、IL-10)的变化。结果与健康对照组相比,模型组大鼠血小板参数PLT、PCT、IL-1β、TNF-α显著升高,IL-10降低(P<0.05或P<0.01)。AA大鼠的血小板参数PLT、PCT与IL-1β、TNF-α、足跖肿胀度、关节炎指数呈正相关,与IL-10呈负相关(P<0.01或P<0.05)。结论佐剂关节炎大鼠PLT、PCT是升高的,且与IL-1β、TNF-α、IL-10、足跖肿胀度、关节炎指数具有正相关性;升高机制可能与致炎因子IL-1β和TNF-α升高,抑炎因子IL-10下降有关。

Objective To observe the changes platelet parameters （platelet count, thrombocytocrit, mean platelet volume, platelet distribution width）, cytokines（ IL-1β, TNF-α, IL-10 ） in adjuvant arthritis rats and their correlation ship. Methods 16 SD masculine rats were averagely divided into normal control group, model group, 8 in each. Model group was intracutaneously injected with 0. 1 ml of Freund＇ s complete adjuvant in the right hindlimb. The changes on platelet parameters （ PLT, PCT, PDW, MPV） and cytokines （ IL-1β, TNF-α, IL-10 ） in two group were observed. Results Compared with normal control group, platelet count, thrombocytocrit, IL-1β, TNF-α of rats were obviously increased, IL-10 was obviously decreased in model group（ P 〈 0.05 或 P 〈0.01 ）. 2. PLT, PCT of AA rats were positively correlated with IL-1β, TNF-α, the toes and ankle＇ swelling, arthritis index （AI）, and IL-10 was negatively correlated with them （ P 〈 0. 05 或 P 〈 0. 01 ）. Conclusions PLT, PCT of adjuvant arthritis rats were increased,and have correlation with IL-1β, TNF-α, IL-10, the toes and ankle＇ swelling, arthritis index （AI）. The mechanism of PLT,PCT increase in adjuvant arthritis rats may be relating to the decrease of IL-1β, TNF-α and the increase of IL-10.
