石蒜碱对人白血病U937细胞的凋亡诱导及作用机制 被引量:9

摘要 分别采用MTT试验和流式细胞仪观察石蒜碱对U937细胞的增殖抑制及凋亡诱导作用;应用Western blot法观察石蒜碱对U937细胞bcl-2、Mcl-1、bcl-xL蛋白表达和poly-(ADP)ribose polymerase(PARP)蛋白剪切的影响。结果石蒜碱可明显抑制U937细胞增殖并诱导细胞凋亡,其细胞增殖的半数抑制浓度(IC50)为2.42μM;石蒜碱可抑制Mcl-1、bcl-xL蛋白表达,对Mcl-1蛋白的抑制作用先于PARP蛋白剪切,对bcl-xL蛋白的抑制发生在PARP剪切片段出现之后。认为石蒜碱可用于治疗异常高表达Mcl-1蛋白的相关肿瘤。
