摘要
以乙酰胺为内标物,丙烯酰胺在10~70g/L范围内,丙烯酰胺/乙酰胺的质量比与其积分峰面积比呈线性关系,相对标准偏差为0.079%;pH值影响酶活性测定结果的准确性,pH值低于2.0时,酶反应终止,pH值越低,丙烯酰胺凝聚为聚丙烯酰胺的量越大,200肛L5.0mol/L的盐酸是适宜量;根据谱图中丙烯腈是否出峰,决定反应体系中发酵液的加入量,酶活性较高时,采用0.5mL的发酵液加入量。
A method to evaluate of the activity of acrylonitrile hydratase based on gas chromatography was developed. It was found that the pH value was an important factor in the determination of the activity of acrylonitrile hydratase. When the pH value was below 2.0, the enzyme activity was inhibited completely.
