摘要
采用紫外线诱变方法,对点青霉9606菌株进行诱变处理,获得了高产葡萄糖氧化酶菌株HW2203。HW2203葡萄糖氧化酶催化β—D-葡萄糖具有高度专一性;酶饱和浓度是5000U/L,在此浓度下,线性范围达8000mg/L,催化底物反应动态平衡点是10000mg/L;在室温下存放12个月,酶活性保存率是90.45%:Cu^2+、Fe^2+、Zn^2+对酶活性均有抑制作用,经透析结果证明,该抑制作用是可逆的。酶试剂盒线性范围大于600mg/100mL;在2~8℃条件下,试剂盒保存12个月线性相关系数等于0.9993,工作液可稳定30d。
Using Penicillium notatum 9606 as starting strain, mutagenized with UV, a mutant HW2203 overproducing GOD was obtained. β-D-glucoses was good for HW2203 GOD with the reactors. Saturation concentration by catalytic reaction of GOD was 5000u/L. The limits linear was 8000mg/L The reactors of catalytic reaction of dynamic equilibrium were 10000mg/L The rest enzyme active was 90. 45% after stored at room temperature for 12 months. The enzyme activity was inhibited by Cu^2+ , Fe^2+ and Zn^2+. The dialysis result showed that the enzymes activity were recovered after meta/ion was removed. The limits linear that the reagents with determine β-D-glucoses of blood serum exceeded 600mg/100mL. The storage life of reagents with determine β-D-glucoses of blood serum was 12 months at 2 - 8℃, R2 = 0. 9993, The steady life of reagents with apply type was 30d.
