摘要
目的筛选川芎贴膏体外透皮实验中的最佳接受液。方法利用TP-5型智能透皮扩散仪,以川芎贴膏中亲水性成分盐酸川芎嗪为考察指标,采用分光光度法,比较不同溶媒系统对川芎贴膏的经皮渗透参数的影响以选择最佳接受液,为体外经皮渗透实验的规范化研究奠定基础。结果生理盐水和pH 7.4磷酸盐缓冲液的透皮速率常数J最大;10%乙醇/生理盐水、10%丙二醇/生理盐水、10%PEG 400/10%乙醇/生理盐水、10%PEG 400/生理盐水的J值依次减少。10%PEG 400/生理盐水透皮时滞最长,pH 7.4磷酸盐缓冲液透皮时滞最短。结论对亲水性成分盐酸川芎嗪而言,生理盐水或pH 7.4磷酸盐缓冲液均可作为其最佳接受液。
Objective To study the receptor fluids of percutaneous permeability of Chuanxiong Adhesive Plaster in vitro and choose a good solution. Methods TP - 5 automatic diffusion apparatus was adopted. Ligustrazine hydrochloride in Chuanxiong Adhesive Plaster was determuined by UV spectroscopy , and the parameters of pereutaneous permeability in various fluids were compared. Results The sequence of rate was:NS 〉 PBS 〉 10% ETOH/NS 〉 10% PG/NS 〉 10% PEG400/10% ETOH/NS 〉 10% PEG400/NS ; the longest lag time was 10% PEG400/NS; the shortest was pH 7.4 PBS. Conclusion Saline or pH7.4 buffered saline solution is recommended for hydrophilic Ligustrazine hydrochloride.
