摘要
目的研究金丝桃素及贯叶金丝桃中金丝桃素提取物对人肝癌细胞株HepG2的体外杀伤效应。方法通过测定胞内荧光强度以确定细胞对金丝桃素的吸收,用显微镜观察和MTT法研究金丝桃素及其提取物对细胞的生长抑制作用。结果光活化的金丝桃素对HepG2细胞有显著的体外杀伤效应,其抑制细胞生长的能力与金丝桃素的浓度、给药与光照的时间间隔以及光照能量密切相关。结论金丝桃素及其提取物能明显抑制HepG2细胞的生长,预示金丝桃素及其提取物在癌症治疗中有良好的开发研究前景。
Objective To investigate the antitumor effect of Hypericin and extract in Hypericum performatum L. on HepG2 cancer cell line of human liver in vitro. Methods The cellular uptake of Hypericin was investigated in HepG2 cells by detecting endocellular fluorescence, and assessed antitumor effect of hypericin and the extract by light microscopy and MTT assay . Results Light - activated Hypericin and the extract had obvious killing effect HepG 2 cell in vitro, and its effects were correlated with the concentration of Hypericin, time interval between administration and irradiation , and also the light energy. Conclusion Hypericin and the extract can obviously decrease the growth of HepG2 cancer cell. Hypericin is a powerful natural photosensitizer and should have a good prospect for further development and investigation.
