摘要
目的研究肺癌细胞株A549培养上清是否能够诱导外周血CD4+CD25+调节性T细胞(简称:Treg细胞)频率增高。方法选择20例健康人并分离其外周血PBMC,设上清组和对照组,上清组用A549培养上清和用PHA(植物血球凝集素)+A549培养上清分别诱导,对照组设空白和单用PHA诱导,用流式细胞仪技术检测两组PBMC不同诱导条件Treg细胞的频率变化。结果用A549培养上清诱导后,上清组Treg细胞的频率与对照组空白孔相比,差异有统计学意义(P<0.05);用PHA+A549培养上清诱导后,上清组Treg细胞的频率与对照组PHA孔相比,差异有统计学意义(P<0.05)。结论肺癌细胞株A549培养上清能够诱导CD4+CD25+调节性T细胞频率增高。
Objective To investigation whether the supernatant from cultured spulmonary carcinoma cell line A549 could induce the frequency of CD4^+ CD25^+ regulatory T cell(Treg)up-regulation. Methods Selected 20 cases health adult and separated peripheral blood mononuclear cells(PBMC), setup supernatant group and control group, supernatant group was induced by A549 supernatant and PHA(phytohemagglutinin)+A549 supernatant, control group setup one blank hole and one PHA induced hole, Flow cytometric analysis was employed for CD4^+ CD25^+ regulatory T cell frequencies in two groups. Results After induced by A549 supernatant,the frequencies of Treg in supernatant group were significantly high than control group's blank hole (P〈0.05); after induced by PHA+A549 supernatant,the frequencies of Treg in supernatant group were also showed equal paces significantly high compared with PHA induced hole of control group(P〈0.05). Conclusion The spulmonary carcinoma cell line A549 could up-regulate the frequency of CD4^+ CD25^+ regulatory T cell in PBMC.
