超滤-紫外可见分光光度法测定钙黄绿素囊泡包封率 被引量:7

摘要 目的:建立携载钙黄绿素的胆固醇琥珀酸酯三羟基氨基甲烷盐囊泡包封率的测定方法。方法:采用超滤法分离囊泡与钙黄绿素;紫外可见分光光度法测定钙黄绿素的含量,检测波长为484nm,计算包封率。结果:钙黄绿素在1.11~11.1mg·L^-1范围内线性良好(r=0.9998);不同截留分子量的超滤膜对钙黄绿素的膜透过率没有影响;利用超滤法可以使囊泡与钙黄绿素得到良好的分离,钙黄绿素的膜透过率在96.7%~97.0%,回收率在96.2%~97.0%,囊泡不能够通过超滤膜。利用该方法测得的钙黄绿素包封率为(37.1±2.4)%。结论:超滤-紫外可见分光光度法可以作为钙黄绿素囊泡包封率的测定方法,胆固醇琥珀酸酯三羟基氨基甲烷盐制备的囊泡可以作为水溶性药物的优良载体。 Objective:To establish a method for the determination of entrapment efficiency of calcein in cholesteryl hemisuccinate Tris vesicles. Methods:Ultrafiltration was applied to separate the free calcein from the vesicles. The assay of calcein was carried out using ultraviolet-visible spectrophotometry and the entrapment efficiency was calculated. The detective wavelength was set at 484 nm. Results:The UV method had good linearity in the range of 1.11 -11.1 mg· L ^- 1 (r =0. 999 8 ). The free calcein was well separated from the vesicles with ultrafiltration method'. There was no obvious difference between the different molecules trapping ultrafiltration membrane in transmissivity of calcein. The calcein transmissivity was between 96.7% - 97.0% and the recoveries with blank vesicles were between 96.2% - 97.0%. The vesicles could not penetrate the ultrafiltration film membrane. The entrapment efficiency of calcein for CHEMS vesicles was ( 37. 1 ± 2.4 ) %. Conclusion : The ultrafiltration UV-VIS method could be used for the determination of entrapment efficiency of calcein vesicles and the CHST vesicle is likely a good carrier for water-solubility drug.
