摘要
目的探讨四种人乳腺癌细胞株(MCF-7,SK-BR-3,T-47D和MDA-MB-231)之间,CCL5表达与其侵袭能力间是否有相关性。方法使用RT-PCR方法和采用CellInva-sionAssayKit分别测定四种人乳腺癌细胞株CCL5表达及其侵袭能力。结果CCL5基因在四种细胞株中均有表达,其中MCF-7细胞株中该基因表达量相对较高,MDA-MB-231和T47-D细胞株次之,SK-BR-3细胞株表达量相对较低。MDA-MB-231细胞株侵袭能力最强,SK-BR-3细胞株次之,MCF-7、T-47-D细胞株侵袭能力弱。结论在四种人乳腺癌细胞株之间,尚未发现CCL5mRNA表达量与其侵袭能力间有明显的线性相关性。该研究为进一步探讨CCL5和乳腺癌的关系提供新方向。
Objective To investigate the correlation between CCL5 expression and the ability of invasion in 4 human breast tumor cell lines ( MCF - 7, SK - BR - 3, T - 47D and MDA - MB - 231). Methods The CCL5 expression in 4 human breast tumor cell lines was assayed by RT -PCR and their ability of invasion was examined by cell invasion assay kit (ECM550). Results CCL5 was expressed in 4 human breast tumor cell lines and CT of CCL5 expression determined by RT - PCR was arranged from smaller to greater: MCF - 7, MDA - MB - 231,T47 - D and SK - BR - 3 cell lines. Accordingly,their ability of invasion was also arranged,but from stronger to weaker: MDA -MB -231 ,SK - BR -3, MCF -7 and T -47 - D cell lines. Conclusion By comparing the CCL5 expression in 4 human breast tumor cell lines to their ability of invasion, although it is pity that significant correlation between them was currently not obtained, the primary experiment may be new way following which causality between CCL5 and breast cancer can be further explored.
