摘要 目的探讨Fhit基因和Survivin基因在膀胱移行细胞癌(TCC)中的表达和意义。方法用免疫组织化学SP法检测49例膀胱TCC组织和10例正常膀胱组织中Fhit蛋白和Survivin蛋白的表达。结果10例正常膀胱组织中Fhit蛋白表达均为阳性,Survivin蛋白表达均为阴性;Fhit蛋白在膀胱TCC中阳性表达率为46.9%(23/49),随恶性程度和临床分期增高而阳性表达减少,差异有统计学意义(P<0.05);Survivin蛋白在膀胱TCC中阳性表达率为57.1%(28/49),随恶性程度和临床分期增高而阳性表达增高,差异有统计学意义(P<0.05);膀胱TCC中Fhit蛋白和Survivin蛋白表达呈负相关(P<0.05)。结论Fhit基因和Survivin基因可能是膀胱TCC发生的晚期分子事件,Fhit蛋白和Survivin蛋白异常表达可作为膀胱TCC的肿瘤标志物。Fhit蛋白可能通过肿瘤凋亡抑制途径发挥作用。 Objective To investigate the expression and clinical significance of Fhit gene and survivin gene in bladder transitional cell carcinoma (TCC). Methods The expression of Fhit gene and survivin gene was detected by S - P immunohistoohemistry in 10 cases of the normal urinary bladder tissues and 49 cases of the bladder TCC tissues. Results All the normal urinary bladder tissues were positive for Fhit protein, but negative for survivin protein. The positive rate of Fhh protein in 49 cases of the bladder TCC tissues was 46.9% with the difference in positive rate between grade Ⅰ and grade Ⅲ being significant ( P 〈 0.05 ). The positive rate of survivin in 49 cases of the bladder TCC tissues was 57.1% with the difference in positive rate between grade Ⅰ and grades Ⅱ,Ⅲ being significant (P 〈0.05 ) ,and the difference in positive rate between Tis - T1 and T2 - T4 being significant too ( P 〈 0.05 ). The expressions of Fhit protein and survivin protein were reversely correlated ( P 〈 0. 05 ). Conclusion Fhit gene and survivin gene may be a late molecular event to the bladder TCC. The expression of Fhit protein and survivin protein may be used as a prognostic marker for the bladder TCC. Fhit protien maybe suppress the development of the bladder TCC by the path of apoptosis.
