摘要
为探讨新生儿细胞免疫功能,应用碱性磷酸酶抗碱性磷酸酶(APAAP)桥联酶标法及双抗体夹心酶联免疫吸附法(ELISA),测定30例足月新生儿及10例早产儿脐血T淋巴细胞亚群及活化的淋巴单核细胞分泌γ-干扰素(IFN-γ)、白细胞介素8(IL-8)的水平,并与正常成人比较。结果表明:①新生儿尤其是早产儿脐血T细胞不成熟,脐血CD+3细胞明显减少,CD+4和CD+8细胞数之和大于CD+3细胞;②活化的脐血单个核细胞生成IFN-γ、IL-8的水平明显低下,且IFN-γ、IL-8的生成与孕周呈正相关。提示新生儿淋巴单核细胞的不成熟及细胞因子的生成低下是新生儿期细胞免疫功能低下的原因之一。
The distribution of T lymphocytes and interferon gamma(IFN γ) and interleukin 8 (IL 8)production by mononuclear cells in cord blood and, for comparison, adult peripheral blood weres tudied. It was shown that immaturities existed in T cell subsets in cord blood, especially in pre mature infants, in that the percentage of CD 3 positive cells was significantly lower than that of the control and the sum of CD 4 and CD 8 positive cells exceeded CD + 3 cells. Cord blood mononuclear cells produced less IFN γ and IL 8 after mitogenic stimulation than did adult mononuclear cells. It was also shown that there was a positive correlation between the concentration of IFN γ, IL 8 secreted by cord blood mononuclear cells and the gestational age. These findings indicated that T cell immaturity and di minished cytokine production might underlie some of the immune deficiencies in neonates.
