摘要
以南薯88等12个抗感黑斑病品种为材料,通过甘薯黑斑病室内抗性鉴定,确定其抗感病程度。通过DNA提取、酶切、PCR扩增、凝胶电泳等系列程序摸索和优化,建立了甘薯黑斑病的AFLP分子标记体系。并用该体系找到了与甘薯抗黑斑病紧密相关的特异性DNA片段,对该片段克隆、测序后发现该DNA片段由320个碱基组成,为进一步分析该片段的特异性和灵敏性为甘薯抗黑斑病分子标记辅助育种奠定了基础。
In this study ,Nanshu 88 and other 11 sweet potato varieties was used as test materials and identified through identification of resistance to black rot in sweet potato. Through extracting DNA,the digestion DNA of sweet potato,PCR amplification and etc,we established the AFLP markers technical system for resistance to black rot of sweet potato, Using this system,we found the species fragment which was clesely related to the resistance of sweet potato black rot, These results provided an useful tool to research the molecular marker of sweet potato black rot.
