应用飞行质谱技术筛选室间隔缺损的血清蛋白标志物 被引量:3

摘要 [目的]应用表面增强激光解吸/电离飞行时间质谱技术筛选单纯性室间隔缺损血清蛋白标志物。[方法]采用病例对照设计,用CM10芯片检测56例单纯性室间隔缺损患儿、85例儿科常见病患儿及23例其他先天性心脏病患儿血清,筛选室间隔缺损血清差异蛋白,并通过构建血清诊断模型分析所筛选差异蛋白区分病例与对照能力。[结果]25个蛋白在3组之间的差异具有统计学意义(P﹤0.05),所构建诊断模型ROC曲线下面积、预测准确率、灵敏度及特异度分别为0.948、94.00%、88.24%、96.97%。[结论]单纯性室间隔缺损患儿、儿科常见病患儿及其他先天性心脏病患儿血清蛋白存在差异,筛选出的差异蛋白能够较好地区分病例与对照,这些蛋白质可能是室间隔缺损的血清标志物。 [Objective] To screen the serum protein biomarkers of isolated ventricular septal defect (VSD) by using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) technology. [ Methods] By ease-control study, CMIO ProteinChip were performed to detect the serum 56 children with VSD. 85 children with common pediatric diseases and 23 children with other congenital heart diseases (CHD), and to select distinct proteins of ISD, and construct the serum diagnosis model to evaluate the ability of distinct proteins for distinguishing the VSD and non-VSD. [Results 25 proteins showed significant different among three groups (P 〈 0.05). By the diagnostic model, the area under receiver operating characteristic curve, accuracy, sensitivity and specificity were 0.948, 94.00% (94/100) , 88.24% (30/34) and 96.97% (64/66), respectively. [Conclusion] The serum proteins showed significant difference among the children with VSD, children with common pediatric diseases and children with other CHD. The distinct proteins selected could distinguish the cases and controls effectively, so these proteins may be the serum protein biomarkers for detecting the VSD.
