急性胰腺炎鼠髓系分化因子88、核因子-κB的表达

EXPRESSION OF MYELOID DIFFERENTIATION FACTOR 88,NUCLEAR FACTOR-ΚB IN ACUTE PANCREATITIS

摘要 [目的]分析急性胰腺炎(AP)小鼠胰腺组织中髓系分化因子88(MyD88)和核因子-κB(NF-κB)的表达变化及其与AP之间的关系。[方法]采用动物实验:以腹腔注射大剂量雨蛙素诱导AP模型;以腹腔注射生理盐水作为对照。用荧光定量逆转录-聚合酶链反应(RT-PCR)对胰腺组织中MyD88和NF-κBmRNA做定量分析。[结果]AP各组MyD88的表达均低于对照组(P﹤0.01)。AP各组之间,除I4组与I2组表达无差异外,I2组表达高于I1组,I12组表达高于I4组(P﹤0.05)。NF-κB的表达,只有I12组与对照组,I12组与I4组的表达有差异(P﹤0.05)。[结论]在AP发展过程中,早期可能以MyD88非依赖性途径为主。随着炎症的发展,MyD88依赖性途径才逐渐发挥作用。 [Objective] To clarify the expression changes of Myeloid differentiation factor 88 (MyD88) and Nuclear factor-kB (NF-kB) in pancreatic tissues of mice with acute pancreatitis (AP) and their relationship with AP. [Methods] By animal experiments, the AP models in mice were induced by intraperitoneal injection of high-dose cerulean; Controls were injected by intraperitoneal injection of normal saline. MyD88 and NF-kB mRNA in pancreatic tissues of mice were quantitative analysis by real-time reverse transcription-polymerase chain reaction (RT-PCR). [Results] The expression of MyD88 mRNA in each AP group was significantly lower than that in control group (P〈 0.01). Among theAP groups, besides the expression showed no significant difference between 14 and 12 group, the expression of 12 group showed significantly higher than that of 11 group, while the expression of 12 group revealed higher than that of 14 group (P 〈 0.05). When compared with control group, the NF-KB mRNA expression of 112 showed significant difference, and significant difference was also found between the NF-kB mRNA expression between 112 versus 14 groups (P 〈 0.05). [ Conclusion] In the progression of AP, MyD88-Independent way was dominant independent way in early stage. With the inflammation progressing, MyD88-dependence has effect on pancreas gradually.
