研究发现,单抗免疫吸附法可清除血浆中的HBV,并有剂量、时间和温度效应关系。每毫升含HBV血浆中加入含7.5mg抗体的吸附剂,反复处理3次之后,所测HBV标志物均转为阴性。抗体吸附法对人血浆中18种主要成分含量或活性。
In order to explore the method for inactivating virus in plasma, the efficacy of antibody adsorption method in inactivating HBV in plasma was observed systematically. The immunoadsorbent used in the experiment was formed by cross link of McAb to HBsAg (IgG 1, ascites titer 1∶50 000) and agarose 4 B bead. The adsorbent was added directly to the plasma containing HBV and, after mixing and adsorbing, removed by filtration. The HBV content in the plasma was measured before and after such treatment. The results showed that McAb adsorption method could inactivate HBV in plasma and the efficacy had relationship with dosage, time and temperature. Treatment of the plasma by addition of the adsorbent containing 7.5 mg Anti-HBsAg McAb per 1 ml of plasma for three times resulted in negative conversion of all HBV markerts tested. The method had no significant influence on the content or activities of the main components of plasma and the antigenicity of the plasma.
