川芎嗪注射液对生理状态下大鼠心室组织中硫化氢分泌的影响

摘要 目的:探讨川芎嗪注射液对大鼠生理状态下心室组织中硫化氢(H2S)分泌的影响。方法:将8只Wistar大鼠左、右心室剪碎后分为4组;正常对照组和川芎嗪低剂量组(1.5μg/ml)、川芎嗪中剂量组(15μg/ml)、川芎嗪高剂量组(150μg/ml),每组n=8。用Krebs液作为孵育液,放入37℃恒温水浴箱中振荡孵育4h后,用放射免疫法测定孵育液中H2S的含量。结果:左心室组织,川芎嗪低剂量组H2S含量(0.17±0.03)和川芎嗪中剂量组H2S含量(0.17±0.03)与对照组H2S含量(0.19±0.03)相比,无统计学意义,川芎嗪高剂量组H2S含量(0.14±0.02)明显低于对照组H2S含量(0.19±0.03)(P<0.01)。右心室组织,川芎嗪低剂量组H2S含量(0.60±0.11)和川芎嗪中剂量组H2S含量(0.53±0.11)与对照组H2S含量(0.64±0.13)相比,无统计学意义,川芎嗪高剂量组H2S含量(0.46±0.09)明显低于对照组H2S含量(0.64±0.13)(P<0.01)。结论:在生理状态下,大剂量川芎嗪具有抑制心室组织释放H2S的作用。 Objective: To explore the effects of ligustrazine on the secretion of hydrogen sulfide in the ventricular tissues of rats under physiological condition. Methods:32 rats were divided into control group, high dose group of ligustrazine( 150μg/nll), medium dose group of ligustrazine(15l.μg/ml) and low dose group of ligustrazine ( 1.5μg/ ml) at random. The left and right ventricuiar tissues were incubated in 37℃ thermostatic water bath for 4 hours with Krebs solution as supernatant. After incubation of the ventricular tissues, the sensitive electrode-based method was used to detect the content of H2S in the supernatant. Results: In the left ventricular tissues, there were on differenceof the H2S level betweenthe 1.5μg/ml hgustrazine group(0.17 ± 0.03), 15μg/ml ligustrazine group(0.17 ± 0.03)and control group. However, compared with control group, the H2S level in hige dose group of ligustrazine decreased obviously( P 〈 0.01 ). In the right ventricular tissues, the H2S level in the 1.5μg/ml ligustrazine groups (0.60 ± 0.11 )and 15μg/ml ligustrazine groups(0.53 ± 0.11 )had no significantly difference from that in the control group(0.64 ± 0.13) ( P 〉 0.05), but the H2S level in the 150μg/ml ligustrazine group(0.46 ± 0.09) was significantly lower than that in the control group(0.64 ± 0.13) ( P 〈 0.01 ). Conclusion: Under physiological condition, high dose of ligustrazine can inhibit the release of hydrogen sulfide in the ventricular tissues of rats.
