摘要 通过向摇瓶中添加Mn^(2+),Zn^(2+),Co^(2+),Mo^(2+),Cu^(2+)5种金属离子,考察各种金属离子对肌苷生产菌的生长、葡萄糖的利用以及肌苷积累的影响。初始培养基中添加Mn^(2+)对产苷有促进作用,而Co^(2+)、Zn^(2+)、Mo^(2+)随着浓度的增加有明显的抑制作用。Cu^(2+)能明显抑制菌体的生长,从而抑制肌苷的生成。针对Mn^(2+)对葡萄糖脱氢酶的活力进行了初步的研究,发现添加Mn^(2+)能促进葡萄糖脱氢酶的活力,葡萄糖在葡萄糖脱氢酶的作用下生成葡萄糖酸,增大了HMP途径的通量,放瓶肌苷含量为14.92 g/L,对照组肌苷含量为11.94 g/L,提高25%。 The effect of five metal ions (Mo^6+ ,Zn^2+ ,Mn^2+ ,Co^2+ ,Cu^2+ ) on the growth of bacterium, glucose consumption and biosynthesis of inosine were studied. The supplement of manganese ion in the basal culture medium helped in the improvement of the inosine production. Growth of bacterium, on the other hand, was restrained with the supplement of cobalt ion, zinc ion, molybdenum ion. It was found that with supplement of copper ion the growth of bacterium was obviously inhibited, and inosine production was also repressed. The effect of manganese ion on the ability of sporulation was investigated. It was also found that with supplement of manganese ions, glucose dehydrogenase activity was increased and thus the flux of HMP was enhanced. The inosine production was improved from 11.94g/L to 14.92g/L. Compared with that of the control group, inosine content was higher by 25%.
