摘要
目的观察含有两个巯基的化合物-二巯基丙磺酸钠(DMPS)在体内外的抗肿瘤作用。方法在体外培养的人肺腺癌A549细胞株中,通过MTT实验,观察不同浓度的DMPS对肿瘤细胞生长繁殖的抑制作用;通过荧光显微镜和细胞凋亡-Hoechst33258染色试剂盒,观察不同浓度的DMPS对肿瘤细胞凋亡的诱导作用。在体内小鼠S180肉瘤模型中,观察不同浓度的DMPS对肿瘤生长的抑制作用。结果体外实验中,终浓度为8 mmol/L和16 mmol/L的DMPS应用48h(P<0.01)和72h(P<0.01)后,显著地抑制肿瘤细胞的生长繁殖;终浓度为8 mmol/L和16 mmol/L的DMPS应用6h和9h后,肿瘤细胞凋亡数量显著地增多(P<0.05,P<0.01)。体内实验中,1.5%DMPS以0.2ml/10g体重腹腔注射,1次/d,连续2周,可显著地降低瘤体重量(P<0.05)。结论高浓度的DMPS显著地抑制体外A549细胞系、体内S180细胞系的生长繁殖。
Objective To investigate the antitumor effect of two thiols-bearing compound-sodium dimercaptopropansulfonate (DMPS)both in vivo and in vitro. Methods The human pulmonary adenocarcinoma cell line A549 cultured in vitro was used to test the effect of DMPS with different concentrations on the growth and proliferation of the tumor cells by means of MTT method, and to test the apoptosis induction of DMPS with different concentrations via fluorescence microscope and with cell apoptosis-Hoechst33258 kit. The inhibitory effect of DMPS with different concentrations on the tumor growth in the model of mice sarcomas 180 was investigated. Results In vitro,DMPS, at the final concentrations of both 8mmol/L and 16mmol/L, significantly inhibited the tumor cell growth and its proliferation when administered 48h( P 〈0.01) and 72h( P 〈0.01) later respectively. The final concentrations of both 8mmol/L and 16mmol/L of DMPS significantly increased the number of tumor cells with apoptotic signs when administered 6h( P 〈0.05) and 9h( P 〈0.01) later respectively. In vivo, 1.5 % DMPS was administered daily intraperitoneally at a dose of 0.2ml/10 weight body for 2 weeks significantly reduced the tumor mass( P 〈 0.05). Conclusion High concentrations of DMPS can significantly inhibit the growth and proliferation of both A549 cell line in vitro and s180 cell line in vivo.
