在全麻手术患者中硫喷妥钠对丙泊酚药动学的影响 被引量:1

摘要 目的研究在全麻手术患者中硫喷妥钠对丙泊酚药动学的影响,并与单用丙泊酚时比较。方法16名全麻手术患者分为2组,分别静注丙泊酚2mg.kg-1或1%丙泊酚和2.5%硫喷妥钠1:1等体积混合液0.2mL.kg-1(相当于丙泊酚1mg.kg-1,硫喷妥钠2.5mg.kg-1),用高效液相色谱法测定丙泊酚血药浓度。用3P97程序拟合计算药动学参数。结果静注丙泊酚的药-时曲线符合三室模型,单用组与合用组的药动学参数:t1/2α分别为(0.8±0.4)和(0.7±0.3)min,t1/2β分别为(10.2±15.6)和(6.5±5.0)min,t1/2γ分别为(113.0±47.2)和(159.0±82.5)min,Vc分别为(0.055±0.022)和(0.052±0.041)L.kg-1,CL分别为(17.4±4.5)和(15.5±8.3)mL.h-1.kg-1,AUC分别为(108.92±31.68)和(88.84±59.08)mg.min.L-1。结论二组患者间丙泊酚主要药动学参数的差异无统计学意义,与文献报道基本一致。 OBJECTIVE To study the effect of thiopental on pharmacokinetics of propotbl in patients undergoing general anesthesia. METHODS 16 Patients undergoing general anesthesia were divided into 2 groups randomly. After intravenous administration of propofol 2 mg· kg^-1 or 1% propofol and 2. 5% thiopental in a ratio of 1:1 volume mixture,the serum concentrations of propofol were measured by high performance liquid chromatography. The pharmacokinetic parameters of propotol were estimated by 3P97 program. RESULTS The serum concentration-time profiles of propotol were discribed by a three compartment model. The pharmacokinetic parameters for two groups were as follows: t 1/2α (0. 8 ± 0.4) and (0.7 ± 0. 3 )min, t 1/2β ( 10.2 ± 15.6) and (6.5 ± 5.0) min, t 1/2γ (113.0 ±47.2) and (159. 0 ±82. 5) min,Vc (0. 055 ±0.022) and (0. 052 ±0. 041) L·kg^-1 ,CL (17.4 ±4. 5) and (15.5±8.3) mL·h^-1· kg^-1 ,AUC (108.92 ±31.68) and (88.84 ±59.08) mg·min·L^-1 ,respectively. CONCLUSION No significant differences were found among the main pharmacokinetic parameters in two groups, which is in good accordance with that of the literature reports.
