阿妥伐他汀对人结肠癌细胞基因表达谱的影响

摘要 目的研究阿妥伐他汀对人结肠腺癌colo-320细胞的反式调节基因,并对结果进行验证。方法以阿妥伐他汀作用于人结肠腺癌细胞株colo-320为实验组,对照组加入PBS,提取总mRNA,逆转录为cDNA,进行基因芯片技术分析;采用Western Blot方法检测阿妥伐他汀对细胞中热休克蛋白70(HSP70)表达的影响。结果实验组细胞19种基因的表达水平上调,24种基因的表达水平下调;实验组HSP70表达水平高于对照组。结论阿妥伐他汀可使人结肠癌细胞的基因表达谱发生改变,可能是他汀类药物防治结肠癌的重要分子机制之一。 Objective To clarify the effect of the expression of atorvastatin on spectra of human colorectal adenecarcinoma colo-320 cells. Methods The human colorectal carcinoma cells of the experimental group were treated with atorvastatin. The control group cells were treated with PBS. High quality mRNA and cDNA were prepared and microarray screening was conducted. The effects of atorvastatin on expression of HSP70 protein in colorectal carcinoma cells were detected by Western blotting assay. Results 19 genes were up-regulated and 24 genes down-regulated in atorvastatin treated colorectal adenecarcinoma cells. The level of HSP70 in atorvastatin-treated groups was higher than that in the control group. Conclusions Atorvastatin can alter the expression profile genes in human colorectal adenecarcinoma colo-320 cells. It may be one of the important molecular mechanisms of atorvastatin in preventing and treating colorectal cancer.
