ET-1在参附注射液预防大鼠房室传导阻滞中的可能作用

Effects of ET-1 in prevention of Shenfu injection against auriculo-ventricular block in rats

目的研究ET-1在参附注射液预防房室传导阻滞(AVB)中的可能作用。方法将48只大鼠随机分为4组(每组12只),分别经尾静脉注射0.9%NS0.045mg/(kg.w)、参附注射液2.5、5、10mg/(kg.w),每日2次,连续5天;用维拉帕米(Ver)制备大鼠房室传导阻滞模型,BL-420生物机能信号系统记录大鼠注射Ver前及注射Ver后2、5分钟时的心率、Ⅲ°AVB发生率和死亡率并测定各组大鼠血浆ET-1的含量。结果与NS组、参附注射液2.5mg/(kg.w)组比较,参附注射液5、10 mg/(kg.w)组能明显降低大鼠心率减慢的程度和死亡率,参附注射液10 mg/(kg.w)组能降低Ⅲ°AVB发生率(P<0.05或P<0.01);此外,参附注射液5、10mg/(kg.第3组w)组血浆ET-1含量增加(P<0.05或P<0.01)。结论参附注射液预防大鼠AVB的作用可能与血清浆ET-1的含量增加有关。

Objective To investigate effects of ET - 1 in prevention of Shenfu injection against auriculo - ventricular block （AVB）. Methods Forty eight rats were randomly divided into four groups （ n = 12）. They were injected respectively normal saline 0. 045mg/（ kg · w） , Shenfu injection 2.5mg/（ kg · w） , 5mg/（ kg · w） , 10mg/（ kg · w） twice a day for five days. AVB models were set up with verapamil in rats. Electrocardiogram was recorded by BL - 420 and the changes of electrocardiograph had been compared. In addition, incidence of Ⅲ° AVB, the death rate of rats, endothelin - 1 （ ET - 1 ） contents of plasma were measured. Results Compared with rats which normal saline 0.045mg/（ kg · w） were injected and rats which shenfu injection2.5mg/（ kg · w） were injected, the heart rate was enhanced rapidly in rats which Shenfu injection5mg/（ kg · w） were injected and rats which Shenfu injection10mL/kg were injected （ P 〈0.05 or P 〈0.01）. Death rate were decreased in rats which Shenfu injection5mg/（ kg · w） were injected and rats which Shenfu injection10mL/kg were injected, Ⅲ° AVB incidences in rats which shenfu injection10mg/（ kg 第 4· · w） were injected. However, the ET - 1 contents of plasma were increased in rats which Shenfu injection5mg/（ kg · w） were injected and rats which shenfu injection10mL,/kg were injected （ P 〈0.05 or P 〈 0.01）. Conclusion Shenfu injection has antagonistic effects on AVB in rats. The prevention may be due to the increase of ET - 1.
