摘要
目的建立同时测定茵山莲颗粒中绿原酸和原儿茶酸含量的方法。方法采用HPLC法。色谱柱为DiamonsiL C18柱(4.6mm×200mm,5μm),流动相为乙腈-质量分数为0.2%的磷酸水溶液(体积比10:90),流速为1.0mL·min^-1,柱温为30℃,检测波长为285nm,进样量为20μL。结果绿原酸与原儿茶酸质量浓度分别在4.80~43.2mg·L^-1、4.08~36.7mg·L^-1内与峰面积呈良好的线性关系,平均回收率分别为101.8%(RSD=1.0%,n=9)和98.5%(RSD=1.4%,n=9)。结论该方法操作简便准确,重复性好,可作为茵山莲颗粒质量控制方法之一。
Objective To develop an HPLC method for the simultaneous determination of chlorogenic acid and protocatechuic acid in Yinshanlian (traditional Chinese medicines) granules. Methods Chromatography was performed on a Diamonsil C18 column (200 mm × 4.6 ram, 5μm) with the mobile phase of acetonitrile- 0.2 % phosphoric acid solution( W: W = 10:90). The detection wavelength was set at 285 nm and the column temperature was 30 ℃. Results The linear calibration curve were obtained in the concentration range of 4.80 - 43.2 mg· L^- 1 for chlorogenic acid and 4.08 - 36.7 mg· L^- 1 for protocatechuic acid. The mean recoveries of chlorogenic acid and protocatechuic acid were 101.8 % (RSD = 1.0 %, n = 9) and 98.5 % (RSD = 1.4 %, n = 9), respectively. Conclusions The method can be used to control the quality of Yinshanlian granules, which provides the advantage of quickness, simplicity and repeatability.
