摘要
目的:研究高剂量1318 nm Nd:YAG激光的生物学效应。方法:用波长为1318 nm的Nd:YAG激光连续照射体外培养的处于对数生长期的人胰腺癌PC-3细胞,分组分别照射0、10、15、20、25、35和50s后,继续培养24h,噻唑蓝比色法(MTT)测定各组吸光度(optical density,OD)值,研究细胞增殖情况。结果:照射剂量在122.3~407.6J/cm2,随着激光照射剂量的提高,PC-3细胞的OD值逐渐降低,且照射15s后差异有统计学意义(P<0.05)。结论:高剂量1318 nm Nd:YAG激光对PC-3细胞具有抑制增殖的作用,且呈剂量依赖性。
Objective:To investigate the biological effect of high intensity 1 318 nm Nd:YAG laser on human pancreatic carcinoma cell (PC-3) proliferation. Methods:Seven groups of PC-3 cells were cultured in 96-well plates for 12 h and then irradiated with 1318 nm Nd:YAG continuous wave laser at different intervals (0, 10, 15, 20, 25, 35, 50 s). The laser power was 1.6W and spot diameter 5 mm (laser dose 0~407.6 J/cm^2). Cells were irradiated once and cultured for the next 24 h. MTT was used to assess the cell proliferation. Results:A significant stimulative effect of 1318 nm laser on PC-3 cell proliferation was found (P〈0.05). Conclusion:The PC-3 cell proliferation might be inhibited by high intensity 1 318 nm Nd:YAG continuous wave laser, which suggested that this laser could be beneficial to patients with pancreatic carcinoma.
