摘要 目的研究4-HPR对肺腺癌细胞A549体外生长的影响及机制的初步探讨。方法MTT法观察4-HPR对肺腺癌细胞株A549的生长抑制作用,流式细胞术观察4-HPR诱导A549细胞凋亡作用和对细胞周期的影响,Western blot分析凋亡相关蛋白表达。结果4-HPR抑制A549的增殖呈剂量依赖性;4-HPR诱导A549细胞凋亡,凋亡过程伴随Bcl-2、Bcl-xS/L表达下降,而Caspase-3、Caspase-9的表达无明显变化。结论4-HPR可明显抑制肺腺癌细胞的增殖,通过降低Bcl-2和Bcl-xS/L促进A549细胞凋亡,为进一步探讨4-HPR治疗肺腺癌的机制提供实验依据。 Objective To investigate the effects of 4-HPR on adenocarcinoma of the human lung cell line A549 in vitro and initial mechanisms for apoptosis. Methods The growth inhibitory effect of 4-HPR on A549 was observed by MTT method, the cell cycle and apoptosis was assayed by flow cytometry, protein expression on apoptosis was de- tected by Western blot. Results 4-HPR inhibited the proliferation of A549 cells in a dose-dependent manner. 4- HPR induced apoptosis,with the decreasing of bcl-2, Bcl-x5/L expression, without marked change of expression of Caspase-3, Caspase-9. Conclusion 4-HPR could inhibit the proliferation of pulmonary adenocarcinoma cell remarkably,induce A549 apoptosis through decreasing Bcl-2, Bcl-x5/L expression, and it provides experimental eviclence to futher investigation of 4-HPR mechanisms in pulmonary adenocarcinoma theraphy.
