霉酚酸对高糖诱导的大鼠系膜细胞增生和高糖诱导的TGF-β、CTGF表达的影响 被引量:1

摘要 目的探讨霉酚酸(MPA)对高糖诱导的大鼠系膜细胞(MC)增生和转化生长因子β(TGF-β)、结缔组织生长因子(CTGF)表达的影响。方法以大鼠MC为受试对象,以不同浓度MPA分别干预24h、48h、72h,噻唑蓝(MTT)比色法检测细胞增生情况。用Western blot法测定干预72h时细胞内TGF-β和CTGF蛋白表达水平。结果MPA能抑制高糖诱导的MC增生,且随浓度升高和时间延长,抑制作用逐渐增强。MPA能抑制高糖诱导的MC内TGF-β和CTGF表达。结论MPA可通过抑制MC增生活化及TGF-β和CTGF表达,从而延缓糖尿病肾病肾小球肥大及肾小球硬化的进展。 Objective To examine the effect of mycophenolic acid(MPA) on mesangial cell(MC) proliferation and on expressions of transforming growth factor-β(TGF-β) and connective tissue growth factor(CTGF ) induced by high glucose. Methods The proliferation activity of mesangial cells incubated for 24h,48h and 72h in different concentrations of MPA was observed by MTT assay. The expressions of TGF-β and CTGF were measured by Western blot. Results MPA inhibited the MC proliferation and expressions of TGF-β and CTGF induced by high glucose. Condusions MPA decreases the release of extracellular matrix by inhibiting the MC proliferation and expressions of TGF-β and CTGF, and subsequently slows the development of glomerulosclerosls in diabetic nephropathy.
