摘要
利用实验室筛选得到的胆固醇氧化酶产生菌青霉TS406进行高产菌株的诱变选育,通过紫外线联合硫酸二乙酯(浓度3%)和紫外线联合亚硝基胍(1.2 mg/mL)两次连续复合诱变获得了一株高产型突变株,其产酶活力从546 U/L提高到了1267 U/L,酶活提高了273%,并利用SPSS软件分析证明了突变株的产酶稳定性。
Use the cholesteroloxidase producting strain penicillium TS406 screened out by our laboratory to mutate,through DES (3%) combine UV mutate we screened out higher productivity strain, then we treated the higher productivity strain with NTG(1.2 mg/mL)combine UV. A mutant, named penicillium N81 showed highest production of cholesteroloxidase was obtained, the enzyme activity was increased from 546 U/L to 1267 U/L (by 273%). We testified stability of the stain penicillium N81 through SPSS analysis. The results showed this mutagenic method gets a good effect.
