石棉诱导AM因子对成纤维细胞作用的研究 被引量:8

摘要 不同浓度的石棉纤维与AM共同培养24h后,取上清液加入WI-38细胞培养,分别用3H-胸腺嘧啶(3H-TdR)掺入法和14C-羟脯氨酸(14-Pro)掺入法观察AM上清液对WI-38细胞增殖和胶原合成的影响,并用氯胺-T法测定成纤维细胞总羟脯氨酸含量。结果显示,石棉诱导的AM上清液能显著增加WI-38细胞增殖和胶原合成。3HTdR掺入的cpm达5205,14C-Pro掺入的cpm为7891,远高于对照组537和830(P<0.01)。经此AM上清液作用的WI-38细胞总羟脯氨酸含量为25.86μg/ml,显著高于TiO2对照组14.79μg/ml(P<0.01)。 The effect of AM supernatants incubated by asbestos on fibroblast was studied. AM was in amedium containing various concentration of asbestos for 24 hrs. The effects of AM supernatants onthe proliferation of WI-38 cell and collagen synthsis were observed by incorperation of 3H-thymidine and 14 C-proline,and the total hydroxy-proline (HOP) content in fibroblasts was determined bythe chloramine-T assay. The results demonstrate that AM supernatant induced by asbestos cansignificantly increase the proliferation of WL38 cell and collagen synthesis. The cpm 52O5 in3H-thyTnidine and the cpm 7891 in the 14 C-proline group were much higher than the cpm 537 and 830in the controls (P<0. 01) respective1y. The HOP content in WI-38 cell acted by AM supernatantsinduced by asbestos was 25. 86μg/ml which was significantly higher than that (14. 79μg/ml) of theTiO2 control (P<0. 01).
