摘要
目的探讨脓毒症时小鼠肺、肝、肾及小肠组织髓样细胞触发受体1(TREM1)表达的改变及其意义。方法雄性昆明小鼠60只,随机分为对照组(10只)、假手术组(10只)、盲肠结扎穿孔(CLP)组(40只),CLP法制备脓毒症模型,CLP后6、12、24和48h处死动物分别留取肺、肝、肾及空肠组织,采用半定量逆转录多聚酶链反应(RT-PCR)法检测TREM1、TNF-αmRNA的表达。结果CLP组6、12、24和48h肺、肝、肾组织TREM1及TNF-αmRNA的表达升高,与对照组比较差异有统计学意义(P<0.05或P<0.01),而空肠组织差异无统计学意义(P>0.05);而且CLP后肝、肺、肾、小肠组织TREM1mRNA表达与TNF-αmRNA水平均呈显著正相关。结论脓毒症时小鼠多器官TREM1基因的表达明显上调,可能与组织炎症反应失控及多脏器功能损害密切相关。
Objective To investigate the expression of TREM-1 and TNF-α mRNA in the lung,hepatic,kidney and intestine tissues in sepsis mice.Methods The sepsis model was made by cecal ligation and puncture(CLP).Sixty male mice were randomly divided into 5 groups :normal control(n=10),sham operation group(n=10),CLP(subdivided into 6,12,24,48 h post CLP,n=10 in each).At serial time points in each group,animals were sacrificed,and the tissues of lung,liver,kidney and intestine were harvested to detect the expression of TREM-1 and TNF-a mRNA by using reverse transcription polymerase chain reaction(RT-PCR).Results The expression levels of TREM-1 and TNF-α mRNA were significantly increased in lung,hepatic and kidney tissues during 6,12,24 h after CLP as compared with control group(P〈0.05 or P〈0.01),while in intestine there was no statistically significant difference(P〉0.05).Meanwhile,tissue TREM-1 mRNA expression was significantly correlated with local TNF-α mRNA expression in liver,lung,kidney and small intestine.Conclusion Sepsis could lead to remarkable up-regulation of TREM-1 mRNA expression in vital organs,which might play an important role in the development of sepsis and subsequent multiple organ dysfunction syndrome.
