摘要
探索了灰树花(Grifola frondosa)GIM5.164深层培养中海藻糖积累的规律。结果表明,以培养基组分玉米粉4%+麸皮浸汁2%+葡萄糖1%,pH3.5及转速为130r/min为适宜灰树花积累海藻糖的培养条件。海藻糖积累于快速生长期之后的稳定期,培养时间到第11天时,菌体内的海藻糖含量最高,达到1.20mg/mL,百分含量为11.48%;将已培养11d的灰树花菌体用37.5℃处理6h后可使海藻糖的含量显著增加,达到1.59mg/mL,百分含量为15.20%;在发酵培养基中添加0.1%~0.2%的花生油作为消泡剂可提高菌体的海藻糖含量;产物海藻糖为浅黄色的晶体。
The regulation of accumulation of trehalose in the submerged culture of Grifola frondosa GIM 5.164 was studied. The strain was cultured in the optimized medium containing corn powder 4%, wheat bran 2%, glucose 1%, and pH3.5 and rotated at a rate of 130 r/min. Trehalose could be accumulated in the stable phase, and the productivity of trehalose is up to maximum when the strain had been cultured for 11 days. The productivity of trehalose was 1.20 mg/mL. Higher trehalose level, which was up to 1.59 mg/mL, could be expected by treating Grifola frondosa at 37.5 ℃ for 6 hours. Groundnut oil added at 0.1% to 0.2% as defoamer could increase the level of trehalose. The buff crystal of trehalose was acquired by drying at -40 ℃ for 12 hours after being centrifugated at a rotation rate of 5000 r/min for 20 minutes.
