摘要
利用AOT(Tween 80)/异辛烷反胶束体系萃取番茄籽蛋白,考察了原料的蛋白含量,讨论了温度、W0、助表面活性剂吐温80用量、浓度、pH值、加料量对蛋白萃取率的影响。实验结果表明较好的工艺条件为:在40℃的超声振荡下,AOT浓度为0.08g/mL、W0为20时,助表面活性剂吐温80和异辛烷的体积比为1∶100、KCl浓度为0.1mol/L、pH值在8.0、加料量为0.02g/mL时蛋白萃取率达到47%。
In the tomato-seed contains 29%-43% protein. How processes into it people's food is a very economic value topic. This paper uses AOT(Tween 80)/iso-octane reverse micelles system extracting tomato-seed protein, Surfactant AOT dissolves in the organic solvent and forms reverse micelles, protein dissolves in reverse micelles, this technology can extract protein economical and convenient, also can keep the activity of protein. This paper discussed content of raw material, the temperature, water content Wo, co-surfactant Tween 80 conent, KCI concentration, pH to protein extraction rate influence. The experimental results indicated the optimum conditions: Under 40 ℃ ultrasound extracting, AOT concentration 0.08 g/mL, according to water content Wo adout 20, according the volume ratio of surface active agent Tween 80 and iso-octane about 1 : 100, according KCI concentration about 0.1 mol/L, pH about 8.0, raw material amount 0.02 g/mL, protein extraction about 47%.
