摘要 [目的]寻求紫花苜蓿叶黄素的最佳提取条件。[方法]以新疆大叶苜蓿为试材,采用微波炉提取法从紫花苜蓿中提取叶黄素。设置提取溶剂、物料比配方、提取时间及微波炉提取火力4个单因素实验和L9(33)正交实验,研究提取叶黄素的影响因素和提取紫花苜蓿叶黄素的最佳组合。[结果]影响叶黄素提取效果的主要因素有提取溶剂、温度、料液比、火力。对水、乙醇和石油醚3种试剂的提取效果和提取液的OD值进行对比分析表明,石油醚提取液的吸收值最大,说明叶黄素易溶于有机溶剂。选择出提取紫花苜蓿叶黄素的最佳条件为:提取试剂为石油醚、物料比1∶50、提取时间3 min、提取火力30 W。[结论]根据单因素实验结果和L9(33)正交实验分析得到提取紫花苜蓿叶黄素的最佳组合为:火力30 W、时间2 min、物料比1∶50。 [ Objective ] The aim of the research was to seek for optimum condition of extracting lutein from alfalfa, [ Method] With Xinjiang Daye alfalfa as tested material, the lutein from alfalfa was extracted by microwave oven extraction method. The single factor experiment of extraction solvent, temperature, material ratio formula,extracting time and extraction firepower and L9(3^3) orthogonal experiment were conduct to study the factors of influencing the lutein extraction and the optimum combination of extracting lutein from alfalfa. [Result] The main factors of influencing the extraction effect were extraction solvent, temperature,material ratio formula and extraction firepower. The comparison analysis on the extraction effects and OD value of extracts of 3 extraction solvents such as water, ethanol and petroleum aether showed that the petroleum aether extract got maximum absorption value, indicating that the lutein was easy to dissolved in the organic solvent. The optimum condition of extracting lutein from alfalfa was as follows : the petroleum aether as extraction solvent, the material ratio of 1:50, the extraction time of 3 min and the extraction firepower of 30 W. [ Conclusion] According to the result from the single factor experiment and L9(3^3) orthogonal experiment the optimum combination of extracting lutein from alfalfa was the extraction firepower of 30 W, the extraction time of 2 min and the material ratio of 1:50.
